He took four wickets in the 19th over of the innings to earn his team an unlikely victory over Pune Warriors, in the 22nd match of the IPL, in Pune.
The 30-year-old also boasts of the unique record of taking three hat-tricks for three different teams.
His first one was for Delhi Daredevils in the inaugural edition of the IPL in 2008, while the second was for Deccan Charges in 2011.
Mishra earlier hit a fluent 30 from 24 balls to rescue Hyderabad after they were struggling at 75 for seven at one stage. His cameo helped Hyderabad get to 119 for eight in their 20 overs.
With the ball, he scripted an unbelievable turnaround as he took four wickets in the penultimate over of the match.
Pune Warriors were favourites till then, with 14 needed from 12 balls and four wickets in hand.
But they self-destructed and left their home fans shell-shocked.
The leg-spinner started his final over with the wicket of Pune captain Angelo Mathews, who was caught at long-off for 20.
Two balls later he trapped Bhuvneshwar Kumar leg before wicket for a duck with a googly.
Rahul Sharma tried to slog it over the leg side but missed completely and was bowled for a first-ball duck as Pune kept losing wickets towards the end.
His dismissal left Pune on the verge of defeat, with just one wicket in hand.
Mishra completed his hat-trick in some style, bowling Ashok Dinda with a googly.
The ball pitched perfectly outside the off-stump,driving Dinda forward before it sneaked through his defences and crashed into the stumps.
That wicket gave Mishra his third hat-trick in the IPL, the first bowler to achieve the landmark.
Yuvraj Singh has two hat-tricks to his credit.
Mishra took four wickets for two runs in final over to finish with superb figures of four for 19 that helped Hyderabad steal a 11-run victory and move to second in the IPL standings with eight points from six games.
